The purpose of this book is to examine analytical HPLC as it is actually used in industry. Rather than focus on the technical issues alone, the book acknowledges that technical issues are inseparably intertwined with nontechnical issues. Managerial and regulatory knowledge, project planning, purchasing, reasoning and presentation of data, teaching skills, legal knowledge, and ethical issues are all integral parts of the day-to-day lives of ordinary scientists. Learning such skills is both essential to working effectively in industry and difficult. For the student, the academic bias toward technical excellence sometimes conflicts with the need for excellence in organizational, teamwork, and leadership skills. There are some excellent works on general and scientific management, but much of that writing is wanting in integrating the theory of management with the realities of the workplace. The present work addresses some of these nontechnical subjects.

The technical aspects of analytical HPLC are the principal focus of this book. The goal is to impart the generalist’s breadth with the specialist’s depth. One would think that it would be easy for an analyst in one industry to transfer his skills to an unrelated industry. Often, it is not so easy. Although there are common threads in the issues involved in industrial processes, there is essential, highly specialized knowledge associated with each manufacturing process. While no book can hope to do justice to all of the aspects of analytical HPLC, it is my hope that this book will be of special service to students just entering industry, to those displaced from positions in one industry seeking to retrain in another, and to those, like myself, who simply enjoy understanding the big picture of how things are made.

Today the worldwide roles of HPLC instruments and supplies amount to over two billion USD, and the market is still expanding further.

This text-books offer little or no help in getting oriented to finding the right one among a half dozen 1/16″ ferrules that look almost the same, but, if an inappropriate one is used in a given fitting, it will be ruined. Dr. Kromidas’s book is a gold mine of useful tips. This practice-oriented book does not fall short of explaining the reasons underlying the problem, and what is just as important, it voices caveat from the consequences of the mistakes one can commit in trying to gain control over the instrument and the separation process.

This book is concise yet rich in practical information, a combination that would be difficult to find in print elsewhere. It helps everybody to be a better practicing chromatographer and may give relief to many who have difficulties in gaining control over the instrument and the chromatographic process at large.

The first manuscript of this textbook was written in 1977 for a training course in high-performance liquid chromatography (HPLC) for laboratory technicians at Berne. Its aim is to show the possibilities and problems associated with modern HPLC. The user of this challenging method needs a broad theoretical and practical knowledge and I hope that this book can impart both. To make things easier—and HPLC can be very easy—the theoretical background is restricted to the minimum.

Since the book is intended just to show the principles of HPLC, chapters dedicated to the separation of different classes of compounds have been omitted. The updates and improvements of this new edition are mainly to be found in details such as new references and technical descriptions which match today’s instrumentation. Four new sections have been written, namely on the shelf-life of mobile phases, the mixing cross, the phase systems in ion chromatography, and on measurement uncertainty. Some equations in the ‘zeroth chapter’, Important and Useful Equations for HPLC, have new numeric values because aporosity of 0.65 is more realistic than 0.8 for chemically bonded phases.

Ultra performance liquid chromatography (UPLC) takes advantage of small 1.7 µm particles operated at elevated pressures to achieve uncompromisable separation speed, resolution, and sensitivity (1-5). In order to tackle advantage of this new technology, it will be necessary to migrate methods from high performance liquid chromatography (HPLC) to UPLC, which can at first appear challenging. However, by selecting the proper column chemistry and dimensions, and properly scaling injection volume, flow rate and gradient conditions (if appropriate), the task of migrating methods is really quite easy and straightforward.
